It depends :
If the handle part is white with a knurled edge then yes , you _have_ to unscrew it before pumping then once it's primed , turn it clockwise until it stops turning else it'll leak in air and often fuel out too .
If the hand part ib lack with rounded , smooth shoulder just pump away but be aware you'll have to pump 50 + times to get it primed .
This is why I prefer the older , white knob typ of primer pumps but , I can't seem to find them anymore (hint) .
BTW : if you have the choice , park facing down hill , this will improve starting and priming both .
